Is 21 days of stay in the country, is still require even he has CNI from UK?

If your fiance has his UK CNI, he has to exchange it forthe local Phils version at the embassy. You then need to register at your local municiple where it takes 10 days to process before you are able to marry. You also need to get CENOMAR from NSO to complete your papers. There are other threads here explaining the new rules, use the search, top right to help you.

No.

If your fiancee acquired his CNI in the UK, he wil need enough time to exchange his CNI at the UK embassy and then get married, including registering. I did it all in just over 2 weeks.

I think you mean how long can he stay in the Philippines. He can stay for 21 days regardless of weather he has a CNI or not. Unless of course he gets a visa to stay longer.
